    多囊卵巢综合征患者外周血Micro15a-5p升高及其影响因素*

      摘要
    • 摘要: 目的 探讨初诊多囊卵巢综合症征( Polycystic ovary syndrome,PCOS) 患者外周血白细胞中miRNA-15a-5p水平与PCOS关系。方法 采用实时定量聚合酶链反应(qRT-PCR)检测PCOS患者外周血白细胞中miRNA-15a-5p表达水平，采用Pearson 或者 Spearman rank 方法分析miRNA-15a-5p与PCOS等一系列变量间的相关性。结果 PCOS组患者中miRNA-15a-5p的表达明显高于健康对照组,纠正年龄以及BMI混杂因素后，PCOS组患者miRNA-15a-5p水平与腰围（Waist circumference、WC）、胰岛素稳态模型（Homeostasis model assessment of insulin resistance，HOMA-IR）、空腹胰岛素（Fasting insulin，Fins)、空腹血糖（Fasting blood glucose，FBG）、餐后2h血糖（2hblood glucose，2hBG）呈正相关，miRNA-15a-5p是PCOS独立影响因素。ROC曲线分析结果提示miRNA-15a-5p可作为PCOS潜在的诊断标志物。结论miRNA-15a-5p与PCOS相关，miRNA-15a-5p可能通过IR参与PCOS的发生以及发展，miRNA-15a-5p可作为PCOS患者糖代谢紊乱早期阶段的潜在的分子标志物。

       

      Abstract: ob＜x＞jective To investigate the relationship between the ex＜x＞pression of miRNA-15a-5p in peripheral blood leukocytes of PCOS patients and the occurrence of PCOS.Methods The ex＜x＞pression of miRNA-15a-5p in peripheral blood leukocytes of PCOS patients was detected by qRT-PCR. The correlation between miRNA-15a-5p and a series of variables were verified by Pearson or Spearman rank analysis. Results The ex＜x＞pression of miRNA-15a-5p in peripheral blood leukocytes of patients in PCOS group was significant higher than that in Control group. The ex＜x＞pression of miRNA-15a-5p in PCOS group was positively correlated with Waist、HOMA-IR、Fins、FBG and 2hBG. Moreover, The ex＜x＞pression of miRNA-15a-5p in peripheral blood leukocytes is an independent influencing factor of PCOS. The results of ROC curve analysis showed that miRNA-15a-5p could be a potential diagnostic biomarker of PCOS.Conclusion: This study reveled the relationship between miRNA-15a-5p and PCOS. miRNA-15a-5p might play an important role in the occurrence and development of PCOS by the affect the IR. miRNA-15a-5p can be used as a potential molecular marker in the early stage of glucose me＜x＞tabolism disorder in PCOS patients.Key Words：PCOS; miRNA-15a-5p; Insulin resistance
