Abstract:
ob<x>jective To explore whether nocardia rhomboides cell wall skeleton (Nr-CWS)can enhance the secretion function of Adipose derived stem cells (ADSCs) to improve the therapeutic effect of diabetic wounds. Methods In vitro, Nr-CWS was used to stimulate ADSCs and cell supernatant was collected (ADSCs-CM) (ADSCs -CM Nr-CWS );The cytokine content in the supernatant was detected by ELISA kit .Human umbilical vein endothelial cells (HUVEC) angiogenesis experiment was conducted to investigate the effect of NR-CWS on angiogenesis. CCK-8、Transwell and scratch experiments were used to detect the effects of ADSCs -CM Nr- CWSon the proliferation and migration of HUVEC . In vivo experiment, the diabetic wound model of mice was made. ADSCs -CM Nr-CWS were injected around the wound surface, and the speed and quality of wound healing as well as the level of wound angiogenesis were analyzed. Results ADSCs -CM Nr-CWS contained more VEGF, bFGF and HGF cytokines than ordinary ADSCS-CM (P<0.05). Compared with ADSCS-CM, ADSCs -CM Nr-CWS could improve cell proliferation, migration and angiogenesis of HUVEC (P < 0.05). The level of wound angiogenesis and wound healing rate of experimental animals treated with ADSCs -CM Nr-CWS were significantly increased (P < 0.05), and the quality of wound repair was also significantly improved. Conclusions Nr-CWS can promote the angiogenesis of diabetic wounds and wound healing by improving the secretion function of ADSCs .
