    Effect of PF429242 on the expression of IL-6 in the brain tissue of mice with cerebral ischemia reperfusion injury

    • 摘要: 目的 研究小分子PF429242对小鼠脑缺血再灌注的保护作用是否与抑制炎症因子白介素-6(IL-6)表达水平有关。方法 将小鼠分为5组,用线栓法制备小鼠大脑中动脉栓塞(MCAO)模型,使用TTC染色法初步评价PF429242对小鼠脑缺血再灌注的保护作用,通过苏木精-伊红染色观察不同脑区细胞的凋亡状态,通过Western blot法评价IL-6的蛋白表达水平。结果 PF429242可减少小鼠脑梗死体积,减少脑细胞的凋亡,降低脑组织IL-6蛋白表达水平。结论 PF429242对小鼠脑缺血再灌注损伤的保护作用可能与其能够抑制小鼠脑缺血再灌注后的炎症反应相关。

       

      Abstract: Objective To investigate whether the protective effect of small molecule PF429242 on cerebral ischemia-reperfusion in mice is related to inhibition of the expression of inflammatory factor IL-6.Methods Mice were divided into five groups. A middle cerebral artery occlusion(MCAO) model was prepared by the thread embolism method. The protective effect of PF429242 on cerebral ischemia-reperfusion in mice was preliminarily evaluated by TTC staining. The apoptosis of cells in different brain regions was observed by H-E staining. The expression of IL-6 was evaluated by Western blot.Results PF429242 reduced infarct volume, brain cell apoptosis, and the expression of IL-6 protein in brain tissue.Conclusions The protective effect of PF429242 on cerebral ischemia-reperfusion in mice is related to its inhibition of inflammatory reaction after cerebral ischemia-reperfusion in mice.
