    The role of DKC1 gene in proliferation and migration of human renal carcinoma cells

    • 摘要: 目的 探讨小干扰RNA (siRNA)沉默DKC1基因对人肾癌细胞株(ACHN、786-O)细胞增殖和迁移能力的影响。方法 分别用对照 siRNA和DKC1 siRNA瞬时转染人肾癌细胞株ACHN和786-O细胞;Western blot实验检测DKC1和EZH2蛋白表达量,细胞增殖实验检测DKC1基因对肾癌细胞增殖能力的影响,Transwell细胞迁移实验检测沉默DKC1基因后对ACHN和786-O肾癌细胞迁移能力的影响。结果 转染DKC1 siRNA后肾癌ACHN及786-O细胞的DKC1表达下调,随着DKC1基因的沉默,EZH2的表达和2种细胞的增殖及迁移能力也下降。结论 沉默DKC1基因后,可以降低EZH2的表达,通过抑制EZH2从而抑制肾癌ACHN细胞及786-O细胞的增殖和迁移能力。

       

      Abstract: Objective To investigate the effects of DKC1 knockdown by small interfering RNA(siRNA) on the proliferation and migration of human renal carcinoma ACHN cells and 786-O cells. Methods The scrambled siRNA and DKC1 siRNA fragments were transiently transfected into ACHN and 786-O cells. The level of DKC1 and EZH2 were detected by Western blot. The effects of DKC1 knockdown on cell proliferation were tested by cell proliferation assay. The effects of DKC1 knockdown on the migration of ACHN and 786-O cells were explored by Transwell assay. Results DKC1 siRNA could down-regulated the level of DKC1 in ACHN and 786-O cells and inhibited the capacity of proliferation and migration of cells via reducing EZH2 expression. Conclusion DKC1 knockdown can reduce the expression of EZH2, which would then inhibit the proliferation and migration of renal carcinoma cells
