    益生菌联合制剂对洛哌丁胺诱导小鼠便秘的影响及机制*

    • 摘要: 目的 建立洛哌丁胺诱导的小鼠便秘模型,使用益生菌联合制剂对模型进行干预,探讨益生菌联合制剂对便秘的改善作用及相关机制。方法 30只小鼠随机分为正常对照组(Normal),便秘组(Constipation)和益生菌组(Probiotic),每组10只;Constipation组和Probiotic组给予洛哌丁胺皮下注射5mg·kg-1,每天两次,连续7天,建立小鼠便秘模型,Normal 组给同体积生理盐水对照。造模成功后,Probiotic组给予益生菌联合制剂mg·kg-1灌胃,其余组给同体积生理盐水对照,连续干预14天后,检测各组小鼠的粪便指标和小肠推进率;石蜡切片观察各组小鼠结肠组织变化;免疫印迹法检测各组小鼠结肠组织中bcl-2,bax蛋白含量表达。结果 与Normal组相比,Constipation组小鼠粪便指标和小肠转运率显著下降,结肠组织中bcl-2/bax蛋白表达比例倒置(<0.05)粘膜层杯状细胞减少,腺体减少,大量炎性细胞浸润;经益生菌联合制剂干预7天后,Probiotic组与Constipation组相比,各组小鼠粪便指标和小肠转运率均显著提高(<0.05),益生菌联合制剂干预14天后小鼠粪便指标和小肠转运率更加显著(<0.05),结肠粘膜损伤,腺体和杯状细胞,炎性细胞浸润结肠组织中bcl-2/bax蛋白表达比例上调(<0.05)。结论 长期使用益生菌制剂可显著改善小鼠的便秘症状,有效结肠损伤这种改善作用可能与上调bcl-2抑制bax的表达,从而抑制细胞凋亡的发生有关。

       

      Abstract: Objective To explore the effect and mechanism of probiotic in treatment of constipation induced by loperamide in ICR mice. Methods 30 ICR mice were randomly divided into three groups: normal group, constipation group, probiotic group , 5 mg·kg-1 of loperamide was given intraperitoneally to constipation group and probiotic group for 7 days to establish the models of constipation. Then the probiotic group were given probiotic 10 mg·kg-1 daily by gavage, for 14 consecutive days. Mice in the normal group, constipation group received an equal volume of saline for 14 consecutive days. At Day 14, the fecal parameters and small intestinal transit were detected. The expression of bcl-2 and bax were determined by Western blotting analysis. Results Compared with the normal group, the fecal pellets, fecal water content, intestinal transit ratio and bcl-2 protein expression decreased in constipation group (P < 0.05),but bax protein expression was significantly increased (P < 0.05); Compared with the constipation group, the fecal pellets, fecal water content, intestinal transit ratio and bax protein expression were significantly reduced in the probiotic group (P < 0.05), but bcl-2 protein expression was significantly increased (P < 0.05). Meanwhile the histological parameters were also improved in the probiotic groups. Conclusion Probiotics can significantly relieve the symptoms of constipation and histological parameters in mice by regulating bcl-2 and bax protein expression ratio
