    TSG101调控p53促进肝癌细胞增殖的机制研究

    TSG101 promotes the proliferation of hepatocellular carcinoma cells by regulating p53

    • 摘要: 目的 探索TSG101调控肝癌细胞增殖的分子机制。方法 在HepG2和SMMC-7721细胞中构建TSG101沉默细胞系及瞬时转染外源TSG101。1、利用western blotting和免疫组化实验分析TSG101在肝癌中的表达；2、利用EdU、细胞集落、CCK-8实验，分析TSG101对肝癌细胞增殖的影响；3、利用western blotting分析TSG101影响肝癌细胞增殖的分子机制。结果 1、TSG101在肝癌中表达增高；2、TSG101能够促进肝癌细胞的增殖；3、TSG101能够负调控p53。结论 1、TSG101是一个促癌因子；2、TSG101可以通过负调控p53促进肝癌细胞的增殖。

       

      Abstract: Objective: To explore the molecular mechanism of TSG101 regulating the proliferation of hepatoma cells. Methods: In HepG2 and SMMC-7721 cells, constructed the stable expression of TSG101 shRNA cells lines and transient transfection of exogenous TSG101. 1. Western blotting and immunohistochemistry were used to analyze the expression of TSG101 in HCC; 2. EdU, cell colony, CCK-8 were used to analyze the effect of TSG101 on the proliferation of hepatoma cells; 3. Western blotting was used to analyze the molecular mechanisms of TSG101 regulating the proliferation of hepatoma cells. Results: 1. The expression of TSG101 is increased in HCC; 2. TSG101 can promote the proliferation of hepatoma cells; 3. TSG101 can negatively regulate the expression of p53. Conclusion: 1 . TSG101 is a cancer promoting factor；2. TSG101 can promote the proliferation hepatoma cells by negatively regulating p53
