    Effects of the pannexin 1 specific inhibitor probenecid on rat with myocardial ischemia reperfusion injury

    • 摘要: 目的探讨丙磺舒特异性阻断泛连接蛋白1(Pannexin1)通道对大鼠心肌缺血再灌损伤的作用。方法免疫组化法确定Pannexin1在大鼠心脏中的表达情况,采用大鼠离体及在体心脏缺血再灌注模型,测定血流动力学指标及离体实验灌注各时间点冠脉流出液和在体实验动物血清中乳酸脱氢酶(lactate dehydrogenase, LDH)含量。实验结束后采用TTC染色测定在体心肌细胞中的梗死面积。结果在心脏组织水平上,Pannexin1主要于心室肌表达。与对照组比较,Pannexin1的阻断剂丙磺舒明显改善左心室发展压,减少在体心肌梗死面积,同时降低心肌细胞LDH的释放。结论Pannexin1在心室肌上表达并在心肌缺血再灌注中起作用,特异性阻断Pannexin1减轻缺血再灌注引起的心肌损伤,起到心肌保护作用。

       

      Abstract: ObjectiveTo investigate the specific effects of pannexin1 on rats with myocardial ischemia reperfusion injury. MethodsThe expression of pannexin1 in rat heart was determined by immunohistochemical method. Then, in vitro and in vivo cardiac ischemia reperfusion models of rats were employed to determine the hemodynamic changes, coronary outflow at each time point of perfusion and the content of serum lactic dehydrogenase (LDH). The amount of formazan in isolated myocardial tissues was measured to determine myocardial activities. The myocardial infracted area was detected by TTC staining. ResultsPannexin1 were mainly expressed in ventricular myocytes. Compared with the group, probenecid, the inhibitor of pannexin1 significantly improved left ventricular end diastolic pressure, enhanced the content of formazan in isolated myocardial tissues, reduced the myocardial infarct area induced by ischemia perfusion and decreased the levels of LDH both in vitro and in vivo. ConclusionPannexin 1 is expressed in ventricular tissue and plays a role in myocardial ischemia reperfusion. Specific blockage of pannexin1 attenuates myocardial injury induced by ischemia reperfusion, which plays a protective role in myocardium.
