摘要:
目的通过观察水通道蛋白4、水通道蛋白9(AQP4、AQP9)在膝骨性关节炎兔模型的膝关节软骨中的表达,从而探讨AQP4、AQP9 在膝骨性关节炎发生发展过程中的作用。方法采用兔膝关节伸直位制动膝骨性关节炎(OA)模型,取制动2、4、6、8和10周的兔膝关节股骨髁软骨组织制作病理切片,在光学显微镜下观察病理改变,并应用免疫组化检测各组关节软骨中AQP4、AQP9的表达。结果随着造模时间延长,兔膝关节软骨破坏程度不断加深;免疫组化染色显示膝骨性关节炎模型兔的膝软骨各时期AQP4、AQP9的表达均高于空白对照组,并且差异有统计学意义(P<0.01)。结论AQP4、AQP9在膝骨性关节炎软骨中高度表达,推测其可能影响骨性关节炎的发生发展。
Abstract:
ObjectiveTo investigate the expression of aquaporin 4 (AQP4) and aquaporin 9 (AQP9) in the knee articular cartilage of rabbits with knee osteoarthritis (OA), so as to discuss the roles of AQP4 and AOP9 in the pathogenesis of knee osteoarthritis. MethodsThe right hind limbs of rabbits were immobilized with a plaster cast in the extension position to establish an OA model. The rabbits were sacrificed after 2, 4, 6, 8 and 10 weeks and their femoral condyle cartilage were taken to prepare histopathology sections for optical microscopic observation. ResultsThe damages in rabbit cartilages were worsen as the modeling process extended. According to immunohistochemistry, the expression of AQP4 and AQP9 were higher in the knee cartilage of the experimental groups than that of the control group, with statistical difference (P<0.01). ConclusionsAQP4 and AQP9 are highly expressed in the knee cartilage of OA, which indicates that AQP4 and AQP9 may be associated with the pathogenesis of OA.
