    庆大霉素对CIH子代大鼠Connexin26蛋白表达水平影响的研究

    • 摘要: 目的 研究宫内缺氧和氨基糖苷类抗生素(庆大霉素)双重影响对仔鼠内耳听力损伤的关系 方法 建立CIH孕鼠模型, SD孕大鼠24只分正常组和缺氧组两组,每组各12只。生后8w,随机选取两组仔鼠再分为四个亚组,每组各12只:称(1)正常仔鼠组(2)正常+GM仔鼠组(3)缺氧仔鼠组(4)缺氧+GM仔鼠组。四组仔鼠做听性脑干反应(ABR)检测听功能后,麻醉处死动物,取出内耳分别做WB、Real-timePCR、甲基化等检查。结果 ①孕鼠动脉血气分析,缺氧组孕鼠动脉PaO2、SaO2水平和正常组比较,均显著下降,差异有统计学意义。但PaCO2及pH值比较,差异均无统计学意义。②ABR检测结果:第(4)组仔鼠ABR反应阈与其它组比较,有明显差异( P < 0 . 01) ;第(2)组和第(3)缺氧仔鼠组两 组 之间 ABR 反 应阈 值差异无统计学意义(P > 0 . 05 ),但和第(1)组ABR反应阈比较,有明显差异( P < 0 . 01) 。③Real Time-PCR 检测仔鼠肝组织GJB2 mRNA 水平和Western Blot 检测仔鼠内耳 Connexin26 蛋白的表达的结果一致,既第(4)组仔鼠GJB2 mRNA 水平与其它组比较,有明显差异( P < 0 . 01) ;第(2)组和(3)组两 组 之间GJB2 mRNA水平差异无统计学意义(P > 0 . 05 ),但和第(1)比较GJB2 mRNA,有明显差异( P < 0 . 01) 。④甲基化测序: GJB2基因启动子岛1、岛2区域甲基化分析, 第(4)组内耳组织中的GJB2基因启动子区域岛1、岛2位点甲基化率百分比明显高其它三组(P<0.05)。第(2)组和第(3)GJB2基因启动子区域岛1、岛2位点甲基化率百分比水平无差异(P > 0 . 05 ),但和第(1)组比较明显升高,有明显差异统计学意义( P < 0 . 05) 结论 妊娠期缺氧及庆大霉素GM的作用可导致仔鼠听力损伤,并且有协同效应。主要是致聋基因GJB2基因启动子岛1、岛2区域甲基化水平上升,GJB2 mRNA水平下降,Connexin26蛋白表达下降。

       

      Abstract: Purpose Study on the relationship between intrauterine hypoxia and aminoglycoside antibiotics (gentamicin) on the development of inner ear dysplasia in neonatal rats. Methods CIH rat model was established. 24 SD pregnant rats were divided into normal group and hypoxia group with 12 rats in each group and 8 weeks after birth. The two groups were randomly divided into four subgroups.( 12 rats in each group) : (1)Normal offspring group,(2) Normal GM group ,(3) Hypoxia group .(4) Hypoxia GM group Four groups of rats were anesthetized and killed after auditory brainstem response (ABR) was measured. The inner ear was examined by WB、Real-time PCR, methylation and so on. Results: ① Analysis of arterial blood gas in pregnant rats: the level of PaO2 SaO2 in pregnant rats in hypoxia group was significantly lower than that in normal group, and the difference was statistically significant.but the Pa CO2 and pH were significantly lower than those in normal group (P < 0.05). ②The difference was not statistically significant. The results of ABR: compared with other groups, there was significant difference in ABR response threshold between the first 4 group and the other groups (P < 0.01).There was no significant difference in ABR response threshold between group 2 (P > 0 05) and group 1 (P > 0 05), but there was no significant difference between group 2 (P > 0 05) and group 1 (P < 0 05). There was significant difference (P < 0.01). There was no significant difference in ABR response threshold between group 2 and group 3 (P > 0.05), But there was significant difference between the ABR reaction threshold and the first group (P < 0.01). ③Real Time-PCR was used to detect the level of GJB2 mRNA in the liver tissues of the offspring, and the Western Blot was used to detect the expression of Connexin26 protein in the inner ear of the offspring, and the results were consistent. There was a significant difference in the level of GJB2 mRNA between the fourth groups compared with the other groups (P < 0.01). There was no significant difference in the GJB2 mRNA level between the second groups and the third groups (P > 0.05), but there was a significant difference compared with the first groups (P < 0.01). ④Methylation sequencing: GJB2 gene promoter island 1 and island 2 region methylation analysis showed that the percentage of methylation rate in group 4 was significantly higher than that in the other three groups (P < 0.05). There was no difference between group 2 and group 3 (P > 0.05).But compared with the first group, the difference was statistically significant (P < 0.05). Conclusion: The effect of hypoxia and gentamicin in pregnancy can lead to hearing damage and synergistic effect . The methylation level of GJB2 gene promoter is increased , the level of GJB2 mRNA decreases , and the expression of Connexin26 protein decreases
