    • 摘要: 目的 探讨DKC1(Dyskerin Pseudouridine Synthase 1, DKC1)基因对人结直肠癌细胞增殖与转移能力的影响。 方法 分别使用Control-siRNA(siCtrl组)和DKC1-siRNA(siDKC1组)瞬时转染人结直肠癌细胞HCT116和DLD1,通过免疫印迹验证DKC1蛋白水平的干扰效果;CCK-8实验及细胞划痕实验检测干扰DKC1基因对结直肠癌细胞HCT116及DLD1增殖能力及迁移能力的影响。结果 免疫印迹结果显示转染DKC1-siRNA后DKC1蛋白水平明显降低;CCK-8及细胞划痕实验结果显示转染DKC1-siRNA后结直肠癌细胞HCT116和DLD1的增殖及迁移能力明显降低。结论 干扰DKC1可以抑制结直肠癌细胞的增殖及迁移能力。

       

      Abstract: Objective To investigate the effect of Dyskerin Pseudouridine Synthase 1 (DKC1) on the proliferation and metastasis of colorectal cancer cells. Methods Human colorectal cancer cells HCT116 and DLD1 were transiently transfected with Control-siRNA (siCtrl group) and DKC1-siRNA (siDKC1 group), and the knockdown effect of DKC1 protein level was verified by Western blot; CCK-8 assay and wound healing assay were used to detect interference DKC1. The effect of DKC1 on the proliferation and migration ability of colorectal cancer cells HCT116 and DLD1 was detected by CCK-8 assay and wound healing assay. Results The results of Western blot showed that the level of DKC1 protein was significantly decreased after transfection of DKC1-siRNA. The results of CCK-8 and cell scratch assay showed that the proliferation and metastasis ability of colorectal cancer cells HCT116 and DLD1 were significantly decreased after transfection of DKC1-siRNA. Conclusions Knockdown of DKC1 can inhibit the proliferation and metastasis of colorectal cancer cells.
