    周围神经损伤后lncRNA的表达谱分析

    Transcriptome study of LncRNAs after peripheral nerve injury

    • 摘要: 目的 通过基因芯片转录组学分析长链非编码RNA(long non-coding RNA, lncRNA)在周围神经损伤(peripheral nerve injury,PNI)与再生过程中的表达变化,进一步深入了解周围神经损伤与再生机制,为周围神经损伤与再生的治疗提供新的途径。方法 使用高通量基因芯片技术,检测小鼠坐骨神经损伤后3、7、14 d较未损伤组远端神经组织中lncRNA和mRNA的表达变化,并通过生物信息学方法筛选出差异lncRNA以及差异lncRNA的靶基因,通过PCR检测其相应的表达量在损伤后不同时间点的变化。结果 坐骨神经损伤后,共有5 354个lncRNA表达发生改变,其中最有可能参与神经再生的关键lncRNA及其靶基因包括:NONMMUG036609-Cd9、NONMMUG014387-Cthrc1、NONMMUG 035232-Ezh2、ENSMUSG00000087366-Jun、SOX2OT-Sox2、NONMMUG010575-Sox4。结论 本研究结果表明,lncRNA在周围神经损伤和再生的过程中表达发生改变,这些差异表达的lncRNA可能参与了周围神经损伤后的再生和修复过程,为临床上治疗周围神经损伤提供新的途径和潜在靶点。

       

      Abstract: Objective To investigate the expression of long non-coding RNAs (lncRNAs) during peripheral nerve injury (PNI) and nerve regeneration through bioinformatics methods, so as to provide potential therapeutic targets for PNI and regeneration. Methods High-throughput gene chips were used to monitor the changes of lncRNAs and their mRNA in distal sciatic nerve tissues of mice 3, 7 and 14 after injury, and the differential lncRNAs and their target genes were screened by bioinformatics methods. PCR was performed to examine the changes of the same indictors after injury over time. Results Significant differences were found in the expression of 5 354 lncRNAs. The lncRNAs and their targeted genes that are more likely to participate in nerve regeneration were NONMMUG036609-Cd9, NONMMUG014387-Cthrc1, NONMMUG 035232-Ezh2, ENSMUSG00000087366-Jun, SOX2OT-Sox2 and NONMMUG010575-Sox4. Conclusions The lncRNAs with differential expression may participate in peripheral nerve injury and nerve regeneration. The current study provides new thoughts and potential targets for clinical treatment of peripheral nerve injury.
