    YAP靶向抑制剂维替泊芬联合放疗抑制脑胶质瘤生长的作用与机制研究

    • 摘要: 目的 研究Yes相关蛋白(YAP)靶向抑制剂维替泊芬(VP)联合放疗对脑胶质瘤细胞增殖与迁移行为的影响及机制.方法 对脑胶质瘤细胞分别用放疗、VP、VP联合放疗3种不同的方法进行处理,应用集落形成实验、CCK-8实验检测细胞增殖情况;应用免疫印迹实验探究VP联合放疗增强放疗效果的机制;应用划痕实验检测细胞迁移情况.结果 放疗或VP处理均能抑制细胞增殖,放疗联合VP处理的细胞增殖速度受到的抑制效果更为显著;VP联合放疗之后,细胞周期蛋白激酶4(CDK4)和磷酸化Rb(p-Rb)的蛋白表达水平显著低于对照组,细胞周期阻滞,增殖减慢;放疗或VP处理均抑制细胞迁移,放疗联合VP处理后,细胞的迁移能力下降更为显著.结论 VP能够明显提高脑胶质瘤的放疗效果;VP联合放疗通过阻断Yes相关蛋白-转录增强结合域(YAP-TEAD)的结合,阻滞细胞周期,从而抑制细胞的增殖.

       

      Abstract: ob<x>jective To study the effect and mechanism of Verteporfin combined with radiotherapy on the proliferation and migration behavior of gliomas. Methods 1. Colony formation assay and CCK8 assay were used to detect the cell proliferation. 2.Western blot was used to explore the Cyr61, CDK4 and p-Rb levels change after VP combined with radiotherapy. 3. Wound healing assay was used to examine the cell migration ability . Results 1. Both radiotherapy and VP treatment inhibited cell proliferation, while radiotherapy combined with VP treatment inhibited cell proliferation even more. 2.After being treated with VP combined with radiotherapy, cell cycle was blocked and proliferation of cells were slowed down. 3.Radiotherapy or VP treatment inhibited cell migration, while radiotherapy combined with VP treatment weakened cell migration ability any more . Conclusion 1.VP can improve the effect of radiotherapy on gliomas. 2.VP combined with radiotherapy blocks the cell cycle, and then inhibits cell proliferation
