    血浆外泌体中Hsa_circ_0004071作为结直肠癌新诊断标志物的研究

    Hsa_circ_0004071 in plasma exosomes as a new diagnostic marker for colorectal cancer

    • 摘要: 目的 筛选结直肠癌(CRC)患者血浆外泌体中差异表达的circRNA,验证其作为CRC新诊断标志物的可行性.方法 选取2019年10月—2020年7月于徐州医科大学附属医院诊治的48例CRC患者血浆样本作为观察组,再选取同期收治的48例非癌症人群血浆样本作为对照组.初步筛选差异circRNA,再进行血浆外泌体提取、总RNA提取、逆转录及RT-PCR定量分析,选取最终目标circRNA进行深入验证.结果 通过生物信息学分析初步筛选出15条差异表达circRNA.采用RT-PCR定量分析发现Hsa_circ_0004071在CRC患者与非癌症对照组血浆外泌体中显著差异表达,诊断性分析结果显示合并灵敏度为77%,特异度为71%,ROC曲线下面积(AUC)为0.7552,表明Hsa_circ_0004071对CRC患者具有较高的诊断效能,且异常表达的Hsa_circ_0004071与CRC TNM分期、远处转移呈正相关.结论 Hsa_circ_0004071在CRC患者血浆外泌体中高表达,且和TNM分期、远处转移相关,可能成为新的CRC诊断标志物.

       

      Abstract: Aim:Screening the differentially expressed circRNA in plasma exosomes of patients with colorectal cancer (CRC) to identify its feasibility as a new diagnostic biomarker for CRC. Method: Selecting the plasma samples of 48 CRC patients admitted to the Affiliated Hospital of Xuzhou Medical University from October 2019 to July 2020 as the observation group, and select the plasma samples of 48 non-cancer patients admitted during the same period as the control group . Preliminary screening of differential genes was carried out through preliminary experiments, and then plasma exosomes extraction, total exosomal RNA extraction, reverse transc<x>ription and RT-PCR quantitative analysis were performed, and the final target genes were selected for in-depth verification. Result: Total of 15 differentially expressed circRNAs were detected by bioinformatics analysis, RT-PCR quantitative analysis found that the target gene Hsa_circ_0004071 was significantly differentially expressed in plasma exosomes of CRC patients and non-cancer control groups. The diagnostic analysis results showed the pooled sensitivity is 0.77 and the pooled specifity is 0.71, the area under the curve curve is 0.7552, indicating that Hsa_circ_0004071 has a high diagnostic efficiency for CRC patients, and the abnormal ex<x>pression of Hsa_circ_0004071 is positively correlated with TNM staging and distant me<x>tastasis in CRC. Conclusion: Hsa_circ_0004071 is highly expressed in plasma exosomes of CRC patients, and it is related to TNM staging and distant me<x>tastasis, it may become a new diagnostic marker for CRC
