    长链非编码RNA LINC00239在人结直肠癌中的表达及其 对结直肠癌细胞HCT116增殖、转移的影响

    Expression of long non-coding RNA LINC00239 in human colorectal cancer and its effects on the proliferation and metastasis of colorectal cancer cell line HCT116

    • 摘要: 目的 探究长链非编码RNA (long non-coding RNA,lncRNA) LINC00239在结直肠癌及其癌旁组织、结直肠癌细胞株及人正常结直肠黏膜细胞株的表达情况,分析LINC00239对结直肠癌细胞HCT116增殖和迁移、侵袭能力的影响。方法 利用GEO及TCGA数据库分析结直肠癌患者的癌和癌旁组织LINC00239的表达情况,应用实时荧光定量PCR(qRT-PCR)法对LINC00239在结直肠癌和癌旁组织中的表达量进行实验验证。利用GEPIA数据库分析LINC00239表达对结直肠癌患者术后无病生存率的影响。CCK8实验和Transwell迁移、侵袭实验分别检测敲低LINC00239对结直肠癌细胞增殖和转移能力的影响。结果 与癌旁组织比较,LINC00239在结直肠癌组织的表达明显上调(P<0.001)。LINC00239低表达组的无病生存期优于高表达组(P=0.033)。与人正常结直肠黏膜细胞株相比,LINC00239在3种结直肠癌细胞株的表达水平明显上调(P<0.05),其中HCT116细胞上调最高(P<0.001)。敲低LINC00239可抑制HCT116细胞的增殖(P<0.001)、迁移(P<0.001)和侵袭(P<0.001)能力。结论 LINC00239在结直肠癌组织和细胞中的表达明显上调,敲低LINC00239可明显抑制结直肠癌HCT116细胞的增殖和转移能力。

       

      Abstract: Objective To investigate the expression of long non-coding RNA (lncRNA) LINC00239 in colorectal cancer and adjacent normal tissues, colorectal cancer cell lines and human normal colorectal mucosa cell lines, and to analyze the effects of LINC00239 on the proliferation, invasion and metastasis of colorectal cancer HCT116 cells. Methods GEO and TCGA databases were used to explore the expression of LINC00239 in colorectal cancer and adjacent normal tissues. The relative levels of LINC00239 in colorectal cancer and adjacent normal tissues were measured by qRT-PCR. GEPIA database was used to analyze the effects of LINC00239 expression on the disease-free survival rate of colorectal cancer patients after surgery. The cell counting kit-8 assay and Transwell assay were used to detect the effects of LINC00239 knocking down on the proliferation and metastasis of colorectal cancer cells. Results Compared with adjacent normal tissues, the level of LINC00239 was significantly up-regulated in colorectal cancer tissues (P<0.001). The low LINC00239 expression group presented better disease-free survival than the high LINC00239 expression group(P=0.033). Compared with human normal colorectal mucosa cell lines, the expression of LINC00239 in three colorectal cancer cell lines was significantly up-regulated (P<0.05), where the expression of LINC00239 was the highest in HCT116 cells (P<0.001). Knocking down of LINC00239 expression significantly inhibited the proliferation (P<0.001), migration (P<0.001) and invasion capability (P<0.001) of colorectal cancer HCT116 cells. Conclusions LINC00239 is remarkably up-regulated in colorectal cancer tissues and colorectal cancer cell lines. Knocking down of LINC00239 can suppress the proliferation and metastasis of colorectal cancer HCT116 cells.
