Abstract:
ob<x>jective To investigate the relative ex<x>pression of long non-coding RNA (lncRNA) LINC00239 in colorectal cancer and their corresponding normal-appearing tissues, colorectal cancer cell lines and normal colorectal epithelial cell lines, and to investigate its effect on proliferation ability and invasion capability of colorectal cancer HCT116 cell. Methods GEO and TCGA databa<x>se was used to explore the ex<x>pression of LINC00239 in CRC.The relative ex<x>pression of LINC00239 in colorectal cancer tissues and their corresponding normal-appearing tissues was assessed by qRT-PCR. GEPIA databa<x>se was used to analyze the relationship between linc00239 ex<x>pression and disease-free survival rate of colorectal cancer patients. Cell counting kit-8 assay and Transwell assay were used to assess the effects of linc00239 knockdown on the proliferation ability and me<x>tastasis capability of colorectal cancer cell. Results Compared with non-cancerous tissues of adjacent colorectal cancer, LINC00239 ex<x>pression was significantly up-regulated in colorectal cancer tissues (P<0.001). The ex<x>pression of linc00239 was related to the disease-free survival rate of colorectal patient(P=0.033). Compared with normal colorectal epithelial cell lines, the ex<x>pression of LINC00239 in three colorectal cancer cell lines was significantly up-regulated (P<0.05), and the ex<x>pression was the highest in HCT116 cells(P<0.001). Knockdown of LINC00239 ex<x>pression significantly inhibited the proliferation (P<0.001),migration (P<0.001) and invasion capability (P<0.001) of colorectal cancer HCT116 cells. Conclusion Linc00239 was up-regulated in colorectal cancer tissues and colorectal cancer cell lines. Knockdown of linc00239 can suppress the proliferation and me<x>tastasis capability of colorectal cancer HCT116 cells.
