Abstract:
ob<x>jective To investigate the mechanism of SIRT1 regulating the proliferation and differentiation of neural stem cells.??Method 1.Cell experiment:??Immunofluorescence and Western blotting techniques were used to observe SIRT1 agonist Resveratrol (RSV), inhibitor Nicotianamine (NA) and sirNA-SIRT1 co-cultured with neural stem cells (24h).??To observe its effects on proliferation and differentiation of neural stem cells and the changes of β-catenin nuclear translocation of neural cells.??2. Animal experiments: Adult male SD rats were randomly divided into Sham operation group (Sham), ischemia/reperfusion group (I/R), solvent control group (DMSO) and administration group. RSV and NA were injected, respectively.??Results RSV promoted the proliferation and differentiation of neural stem cells into neurons, while NA inhibited the differentiation of neural stem cells into astrocytes and oligodendrocytes, but RSV and NA did not affect the differentiation of neural stem cells into astrocytes and oligodendrocytes.??It was found that appropriate RSV could promote β-catenin to the nucleus, while SIRT1 siRNA could reverse RSV.??After cerebral ischemia/reperfusion injury, appropriate RSV can promote β-catenin to the nucleus, while NA can inhibit this effect.??Conclusion SIRT1 regulates the proliferation and differentiation of neural stem cells by regulating the nuclear translocation of β-catenin. ?
