    三种茶叶茶多酚提取物的体外抗氧化及其对过氧化氢诱导的RAW264.7细胞氧化应激损伤的保护作用研究

    Study on the Antioxidant Effects of Tea Polyphenols Extracts from Three Kinds of Tea and Their Protective Effects on Hydrogen Peroxide-induced Oxidative Stress of RAW264.7 Cells

    • 摘要: 目的:探究毛尖、铁观音、普洱三种茶叶的水提物的体外抗氧化作用及对H2O2诱导的RAW264.7细胞损伤的保护作用。方法:用福林酚法测定三种提取物中茶多酚含量。采用1,1-二苯基-2-三硝基苯肼(DPPH)法和铁离子还原(FRAP)法对三种茶多酚提物的体外抗氧化活性进行研究。采用噻唑兰比色法(MTT)研究三种茶多酚提物对过氧化氢诱导的巨噬细胞 RAW264.7 细胞损伤的保护作用。结果:毛尖、铁观音、普洱茶多酚提取物的茶多酚含量分别为46.79±0.99%、46.22±1.03%、33.72±0.60%。毛尖、铁观音、普洱各自水提物的DPPH自由基半数清除浓度(IC50)分别为4.33、4.91、13.49 μg/mL 。毛尖、铁观音、普洱水提物的FRAP值分别为350.89±3.11、261.55±2.98、109.97±3.25mmol/L。三种茶叶的水提物在浓度低于80ug/mL时,均对H2O2诱导RAW264.7细胞氧化应激有良好的保护作用。当浓度超过80ug/mL时,细胞存活率随着浓度的上升而下降,呈显著负相关。结论:茶多酚提取物的DPPH自由基清除能力及FRAP值的大小关系均为:毛尖>铁观音>普洱。提取物的抗氧化活性也与茶多酚含量成显著正相关。三种茶叶的水提物在80ug/mL浓度以内均对过氧化氢诱导的细胞RAW264.7氧化应激有保护作用。

       

      Abstract: ob<x>jective : Explore the in vitro antioxidant effects of water extracts of Maojian tea, Tieguanyin tea , Purer tea and their protective effects on hydrogen peroxide(H2O2)-induced of RAW264.7 cells. Methods: The contents of tea polyphenols in the three extracts were determined by Folin colorimetry method. The 1,1- diphenyl -2- trinitrophenylhydrazine (DPPH) method and the total antioxidant capacity (FRAP) evaluation method were used to evaluate the in vitro antioxidant activities of three tea polyphenols extracts. The protective effects of three tea polyphenols extracts protective effects on hydrogen peroxide(H2O2)-induced of Macrophages -RAW264.7 cells were studied by MTT assay. Results: The contents of tea polyphenols in Maojian tea, Tieguanyin tea and Purer tea polyphenols extracts were 46.79±0.99%, 46.22±1.03% and 33.72±0.60%, respectively. The DPPH free radical scavenging concentrations (IC50) of the water extracts of Maojian tea, Tieguanyin tea, and Purer tea were 4.33, 4.91 and 13.49μg/mL,respectively. In the hydroxyl radical experiment, the IC50 of the water extracts of Purer tea, Maojian tea and Tieguanyin tea were 1.02, 1.51 and 1.81 mg/mL, respectively. The FRAP values of the water extracts of Maojian tea, Tieguanyin tea and Puer tea were (350.89±3.11),(261.55±2.98) and (109.97±3.25) mmol/L, respectively.The water extracts of three kinds of tea have good protective effects on hydrogen peroxide(H2O2)-induced oxidative stress of RAW264.7 cells when the concentration is lower than 80ug/ml. When the concentration exceeds 80ug/ml, the cell survival rate decreases with the increase of the concentration, showing an obvious negative correlation. Conclusion: The DPPH free radical scavenging ability and FRAP value of tea polyphenols extract are Maojian tea > Tieguanyin tea > Purer tea. The antioxidant activity of the extract is also positively correlated with the tea polyphenol content. The water extracts of three kinds of tea have protective effects on hydrogen peroxide(H2O2)-induced oxidative stress of RAW264.7 cells.
