    The expression and clinical significance of neuropilin-1 in triple negative breast cancer

    • 摘要: 目的 探讨Neuropilin-1(NRP-1,神经纤毛蛋白-1)在三阴性乳腺癌中的表达及其与临床病理特征的相关性。方法 筛选并回顾分析手术切除治疗的69例三阴性乳腺癌及30例乳腺良性疾病病例,将相应患者病理标本制作成组织标本,并采用免疫组化方法检测三阴乳腺癌组织和乳腺良性疾病组织中NRP-1的表达情况,对NRP-1的表达与三阴性乳腺癌患者的临床病理特征进行相关性分析。结果 ①NRP-1在三阴性乳腺癌中的阳性表达率明显高于乳腺纤维腺瘤和乳腺囊性增生症(P=0.000);NRP-1在乳腺纤维腺瘤和乳腺囊性增生症中的表达无明显差异(P=0.6767);②NRP-1表达率随三阴性乳腺癌术后分期的升高逐渐升高(P=0.000);③三阴性乳腺癌中,腋窝淋巴结转移患者中NRP-1表达率高于无淋巴结转移者(P=0.001);④三阴性乳腺癌患者NRP-1的表达受肿瘤大小、细胞分化程度、pTNM分期、随访期间是否有复发或远处转移影响。结论 NRP-1的过表达可能与三阴性乳腺癌的发生、发展、腋窝淋巴结转移密切相关,并有可能成为三阴性乳腺癌治疗的新靶点。

       

      Abstract: Objective To explore the expression of neuropilin-1 (NRP-1) in triple negative breast cancer and its correlation with clinicopathologic features. Methods A total of 69 triple negative breast cancer patients and 30 patients with benign breast lesions were included into the current study. Their clinical data were retrospectively analyzed, while their pathological specimens were prepared into tissue samples. The expression of NRP-1 was detected by immunohistochemistry in triple negative breast cancer and breast benign lesions. Meanwhile, the relationship between NRP-1 expression and the patients’ clinicopathological features was analyzed. Results The positive expression rate of NRP-1 was significantly higher in triple negative breast cancer than those in breast fibroadenoma and breast cystic hyperplasia (P=0.000). There was no significant difference as to the expression of NRP-1 between fibroadenoma and breast cystic hyperplasia (P=0.6767). The expression rate of NRP-1 was gradually increased as the stage of triple negative breast cancer was elevated after surgery (P=0.000). For triple negative breast cancer patients, those with axillary lymph node metastasis showed a high expression rate of NRP-1 than those without lymph node metastasis (P=0.001). The expression of NRP-1 in triple negative breast cancer patients was associated with tumor size, cell differentiation, pTNM stage and recurrence or distant metastasis in triple negative breast cancer. Conclusions NRP-1 over-expression may be closely associated with the pathogenesis and axillary lymph node metastasis of triple negative breast cancer, and may become a new target for the treatment of the cancer.
