    Intestinal flora in patients with IgA nephropathy — A case-control study

    • 摘要: 目的 通过16S rDNA测序技术探索IgA肾病患者与健康人群肠道菌群构成的差异。方法 选取原发性IgAN患者10例,和年龄、性别和肾功能均衡的社区健康人群10例行病例对照研究。采用16S rDNA技术基于Illumina NovaSeq测序平台,进行物种注释及丰度分析,比较两组样本菌群组成和结构。结果 IgA肾病组与健康对照组在肠道菌群组成上具有显著差异。门分类水平上,IgA肾病组的厚壁菌门相对丰度较健康对照组显著增加,拟杆菌门和放线菌门的相对丰度较健康对照组显著降低。属分类水平上,IgA肾病组中巨单胞菌属的数量较健康对照组显著增加,双岐杆菌属的数量较健康对照组显著降低。结论 原发性IgA肾病患者肠道菌群较健康人群发生显著变化,可能是IgA肾病潜在致病机制。

       

      Abstract: objective To explore the difference of intestinal flora composition between IgA nephropathy ( IgAN) patients and healthy control by 16S rDNA sequencing technology. Methods Ten patients with primary IgAN and 10 healthy people with balanced age, gender, and renal function were selected for a case-control study. Species annotation and abundance analysis were carried out using 16S rDNA technology based on Illumina NovaSeq sequencing platform, and the flora composition and structure of the two groups of samples were compared. Results The composition of intestinal flora was significantly different between the IgAN group and the healthy control. At the level of phylum classification, the relative abundance of firmicutes increased significantly in the IgAN group compared with the healthy control, while the relative abundance of bacteroidetes and actinomycetes decreased significantly compared with the healthy control. On the level of classification, the number of megomonas in the IgAN group increased significantly compared with the healthy control, while the number of bifidobacterium decreased significantly compared with the healthy control. Conclusion The intestinal flora in patients with primary IgAN had significant changes when comparing with healthy people, which might be the underlying pathogenesis of IgA nephropathy.
