Abstract:
ob<x>jectiveTo investigate the effect of CUX1 gene on the proliferation and me<x>tastasis of colorectal canc er.Methods The ex<x>pression of CUX1 in cancer and adjacent tissues of colorectal cancer patients was analyzed by using TCGA databa<x>se, and the ex<x>pression of CUX1 in colorectal cancer tissues was detected by real-time fluorescence quantitative PCR( qRT-PCR).The GEPIA databa<x>se was used to analyze the effect of CUX1 ex<x>pression on the overall survival rate of patients with colorectal cancer after surgery. siRNA-Ctrl and siRNA-CUX1 were transfected into human colon cancer cells.C CK-8 and Transwell assay were used to detect the effect of CUX1 on the proliferation and me<x>tastasis of colorectal cancer cells.The interference efficiency of CUX1 and the ex<x>pression of E-cadherin, Vimentin and N-cadherin proteins were detected by western blotting.Results The results of TCGA databa<x>se and qRT-PCR showed that CUX1 was highly expressed in colorectal cancer tissues (P<0.05).The results of GEPIA databa<x>se showed that the overall survival rate of the low ex<x>pression group was significantly better than that of the high ex<x>pression group (P<0.01).The results of Transwell assay showed that siRNA- CUX1 reduced the me<x>tastatic ability of colorectal cancer cells.CCK-8 assay showed that siRNA- CUX1 inhibited the proliferation of colorectal cancer cells (P<0.001).Western blot results showed that the ex<x>pression of CUX1, N-cadherin and Vimentin in colorectal cancer cells were decreased after siRNA- CUX1 transfection.The ex<x>pression of E-cadherin was increased (P<0.05). Conclusion Knockdown of CUX1 can inhibit the proliferation and me<x>tastasis of colorectal cancer.
