    KLF16在卵巢癌组织中的表达及对卵巢癌细胞生物学行为的影响

    Expression of KLF16 in ovarian cancer and its effect on biological behavior of ovarian cancer cells

    • 摘要: 目的 明确Krüeppel样转录因子16(KLF16)在卵巢癌组织中的表达情况及其对卵巢癌细胞增殖、迁移及侵袭能力的影响。方法 通过TCGA数据库分析KLF16在卵巢癌组织中的表达情况以及KLF16表达水平与卵巢癌患者预后的关系。采用免疫组化及RT-qPCR检测卵巢癌组织及正常卵巢组织中KLF16表达情况。使用Western blot检测卵巢癌细胞系中KLF16的表达情况及转染后SKOV3细胞系KLF16表达下调效果。通过CCK-8、EdU、集落形成实验及Transwell实验验证KLF16对卵巢癌细胞增殖、迁移、侵袭能力的影响。通过Western blot实验检测KLF16对上皮钙黏蛋白(E-cadherin)、神经钙黏蛋白(N-cadherin)及锌指蛋白Snail表达的影响。结果 TCGA数据库显示KLF16在卵巢癌组织中高表达,并且高表达的患者预后较差(P<0.05)。免疫组化及RT-qPCR结果显示卵巢癌组织中KLF16的表达水平高于正常卵巢组织,差异有统计学意义(P<0.05)。KLF16阳性表达与卵巢癌患者的FIGO分期、组织分化程度有关(P<0.05)。SKOV3细胞敲低KLF16后,细胞的增殖、迁移及侵袭能力明显下降,上皮间质转化(EMT)相关蛋白N-cadherin及Snail的表达明显下降,E-cadherin的表达明显增加。结论 KLF16在卵巢癌组织中高表达,且可增强卵巢癌SKOV3细胞增殖、迁移及侵袭的能力。

       

      Abstract: Objective To investigate the expression of Krüppel-like transcription factor 16 (KLF16) in ovarian cancer tissues and its impact on the proliferation, invasion, and migration abilities of ovarian cancer cells. Methods The expression of KLF16 in ovarian cancer tissues and its correlation with patient prognosis were analyzed by the TCGA database. Immunohistochemistry and real-time quantitative reverse transcription polymerase chain reaction (RT-qPCR) were used to detect the expression of KLF16 in ovarian cancer tissues and normal tissues. Western blot was employed to detect the expression of KLF16 in ovarian cancer cell lines and the effect of KLF16 knockdown in SKOV3 cells. The impact of KLF16 on the proliferation, migration, and invasion of ovarian cancer cells was detected by CCK-8, EdU, colony formation, and Transwell assays. Western blot was used to examine the effect of KLF16 on the expression of epithelial cadherin (E-cadherin), neural cadherin (N-cadherin), and zinc finger protein Snail. Results TCGA database analysis showed that KLF16 was highly expressed in ovarian cancer tissues, and patients with high KLF16 expression showed poorer prognosis (P<0.05). Immunohistochemistry and RT-qPCR results revealed that KLF16 expression in ovarian cancer tissues was significantly higher than in normal tissues (P<0.05). The positive expression of KLF16 was associated with the FIGO stage and histological differentiation of ovarian cancer patients (P<0.05). After KLF16 knockdown in SKOV3 cells, the cells showed significant decreases in proliferation, invasion, and migration abilities, with decreased expression of epithelial-mesenchymal transition (EMT)-related proteins N-cadherin and Snail and increased expression of E-cadherin. Conclusions KLF16 is highly expressed in ovarian cancer tissues and can enhance the proliferation, migration, and invasion abilities of ovarian cancer SKOV3 cells.
