    DKC1基因对结直肠癌细胞增殖与转移能力的影响

    The effect of DKC1 gene on the proliferation and metastasis of colorectal cancer cells

    • 摘要: 目的 探讨DKC1基因对人结直肠癌细胞增殖与转移能力的影响。方法 分别使用Control-siRNA(siCtrl组)和DKC1-siRNA(siDKC1组)瞬时转染人结直肠癌细胞HCT116和DLD1,通过免疫印迹验证DKC1蛋白水平的干扰效果;CCK-8实验、划痕实验、Transwell细胞侵袭实验检测干扰DKC1基因对结直肠癌细胞HCT116及DLD1增殖能力及转移能力的影响。结果 免疫印迹结果显示转染DKC1-siRNA后DKC1蛋白水平明显降低;CCK-8实验结果显示转染DKC1-siRNA后结直肠癌细胞HCT116和DLDI的增殖能力明显降低(P<0.01);划痕实验及Transwell细胞侵袭实验结果显示转染DKC1-siRNA后结直肠癌细胞HCT116和DLD1的转移能力明显降低(P<0.05)。结论 干扰DKC1可以抑制结直肠癌细胞的增殖及转移能力。

       

      Abstract: Objective To investigate the effect of dyskerin pseudouridine synthase 1(DKC1)on the proliferation and metastasis of colorectal cancer cells. Methods Human colorectal cancer cells HCT116 and DLD1 were transiently transfected with Control-siRNA(siCtrl group)and DKC1-siRNA(siDKC1 group),and the knockdown effect of DKC1 protein level was verified by Western blot.The effect of DKC1 on the proliferation and metastasis ability of colorectal cancer cells HCT116 and DLD1 was detected by CCK-8 assay,wound healing assay and Transwell assay. Results The results of Western blot showed that the level of DKC1 protein was significantly decreased after transfection of DKC1-siRNA.The results of CCK-8 assay showed that the proliferation ability of colorectal cancer cells HCT116 and DLD1 were significantly decreased after transfection of DKC1-siRNA(P<0.01).Wound healing assay and Transwell assay showed that the metastasis ability of colorectal cancer cells HCT116 and DLD1 were significantly decreased after transfection of DKC1-siRNA(P0.05).Conclusions Knockdown of DKC1 can inhibit the proliferation and metastasis of colorectal cancer cells.
