    长链非编码RNA UCA1在乳腺癌中的作用及机制研究

    Study on the role and mechanism of lncRNA UCA1 in breast cancer cells

    • 摘要: 目的 探讨长链非编码RNA(lncRNA)尿路上皮癌胚抗原1(UCA1)在乳腺癌细胞中的表达及其对乳腺癌细胞增殖能力的影响。方法 采用荧光定量聚合酶链反应(qRT-PCR)检测乳腺癌细胞株BT-459、SKBR3、MCF-7、T47D和MDA-MB231中lncRNA UCA1的表达,通过慢病毒转染构建过表达UCA1乳腺癌细胞株,采用MTT法和流式细胞术检测过表达后乳腺癌细胞增殖能力和细胞周期分布的变化,进一步采用qRT-PCR和Western blot技术检测WNT信号通路中WNT6的表达。结果 与正常人乳腺上皮细胞相比,LncRNA UCA1在乳腺癌细胞株中高表达(P<0.05)。过表达UCA1促进了细胞增殖(P<0.05),并且促进了细胞周期进展(P<0.05)。过表达UCA1促进了WNT信号通路中WNT6基因及蛋白的表达(P<0.05)。结论 UCA1在乳腺癌细胞株中高表达,过表达UCA1后促进了细胞增殖,该作用可能与WNT信号通路相关,本研究为LncRNA的临床应用提供了一定的实验依据。

       

      Abstract: Objective To explore the expression of long non-coding RNA urothelial cancer-associated 1 (lncRNA UCA1) in breast cancer cells and its effect on the proliferation of breast cancer cells. Methods The expression levels of lncRNA UCA1 were analyzed in breast cell lines of BT-459, SKBR3, MCF-7, T47D and MDA-MB231 through quantitative reverse-transcription polymerase chain reaction (qRT-PCR) analysis. The lentivirus UCA1 was transfected into breast cancer cells. The cell proliferation ability and cell cycle distribution were detected by flow cytometry and MTT assay respectively. The expression of WNT6 in WNT signaling was examined by qRT-PCR and Western blot. Results The expression levels of lncRNA UCA1 were higher in breast cancer cell lines compared with normal breast epithelial cell (P<0.05). Overexpression of UCA1 significantly enhanced cell proliferation and cell cycle progression (P<0.05). And both gene and protein level of WNT6 were improved after overexpression of UCA1 (P<0.05). Conclusions lncRNA UCA1 is highly expressed in breast cancer cell lines. Overexpression of UCA1 promotes cell proliferation, which may be related to WNT signaling pathway. This study provides some experimental basis for the clinical application of lncRNA.
