Abstract:
Objective To investigate the effects of
KIF4A gene on the migration and invasion of colorectal cancer cells and related mechanisms.
Methods The
KIF4A-CtrlRNA (si-Ctrl) and
KIF4A-siRNA (si-KIF4A) were transiently transfected into colorectal cancer cell lines SW480 and DLD1, and the interference effect was verified by Western blot. The effects of KIF4A on the migration and invasion ability of colorectal cancer cells were confirmed with Transwell assay. The changes of migration and invasion-related proteins MMP2 and MMP9 were detected by Western blot.
Results Compared with si-Ctrl group, the si-KIF4A group presented remarkable decreases in KIF4A protein levels, cell migration and invasion abilities, and the levels of MMP2 and MMP9 (
P<0.001).
Conclusions KIF4A facilitates the migration and invasion abilities of colorectal cancer cells through stimulating the expression of MMP2 and MMP9.
