    多囊卵巢综合征患者外周血miRNA-15a-5p升高及其影响因素

    Increases of miRNA-15a-5p level in the peripheral blood of PCOS patients and the influencing factors

    • 摘要: 目的 探讨初诊多囊卵巢综合征(polycystic ovary syndrome,PCOS) 患者外周血白细胞中miRNA-15a-5p水平与PCOS关系。方法 采用实时定量聚合酶链反应(qRT-PCR)检测PCOS患者外周血白细胞中miRNA-15a-5p表达水平,采用Pearson 或者 Spearman rank 方法分析miRNA-15a-5p与PCOS等一系列变量间的相关性。结果 PCOS组患者中miRNA-15a-5p的表达明显高于健康对照组,纠正年龄以及体重指数(BMI)混杂因素后,PCOS组患者miRNA-15a-5p水平与腰围(waist circumference,WC)、胰岛素稳态模型(homeostasis model assessment of insulin resistance,HOMA-IR)、空腹胰岛素(fasting insulin,FINS)、空腹血糖(fasting blood glucose,FBG)、餐后2 h血糖(2 h blood glucose,2hBG)呈正相关,miRNA-15a-5p是PCOS独立影响因素。ROC曲线分析结果提示miRNA-15a-5p可作为PCOS潜在的诊断标志物。结论 miRNA-15a-5p与PCOS相关,miRNA-15a-5p可能通过胰岛素抵抗参与PCOS的发生以及发展,miRNA-15a-5p可作为PCOS患者糖代谢紊乱早期阶段潜在的分子标志物。

       

      Abstract: Objective To investigate the relationship between the expression of miRNA-15a-5p in peripheral blood leukocytes of polycystic ovary syndrome (PCOS) patients and PCOS. Methods The expression of miRNA-15a-5p in peripheral blood leukocytes of PCOS patients was detected by qRT-PCR. The correlation between miRNA-15a-5p and a series of variables were analyzed by Pearson or Spearman rank analysis. Results The PCOS group showed significant higher levels of miRNA-15a-5p than the healthy control group. After correction of age and BMI, the level of miRNA-15a-5p in the PCOS group was positively correlated with waist circumference (WC), homeostasis model assessment of insulin resistance (HOMA-IR), fasting insulin (FINS), fasting blood glucose (FBG) and 2-hour blood glucose (2hBG). MiRNA-15a-5p is an independent influencing factor of PCOS. According to ROC curve analysis, miRNA-15a-5p could be a potential diagnostic biomarker of PCOS. Conclusions MiRNA-15a-5p is associated with PCOS. MiRNA-15a-5p may involve the pathogenesis of PCOS through insulin resistance. MiRNA-15a-5p can be used as a potential molecular marker in the early stage of glucose metabolism disorder in PCOS patients.
