自噬调控肿瘤放疗敏感性的研究进展

摘要: 放疗是肿瘤的常用治疗手段之一,但肿瘤细胞的放疗抵抗性会极大地降低放疗疗效,造成抗肿瘤治疗的失败,引起肿瘤的复发和转移。自噬作为细胞内普遍存在的一种代谢途径,对于维持细胞内稳态具有重要作用。研究表明,自噬可以发挥肿瘤促进和肿瘤抑制的双重作用。一方面,自噬使肿瘤能够适应缺氧和营养不足等应激环境,增强肿瘤细胞的耐受性,维持细胞生存;另一方面,长期的过度自噬也可导致肿瘤细胞的自我降解,引起细胞死亡。本文综述了近年来关于自噬调控肿瘤放疗敏感性具体机制的前临床研究,旨在为通过靶向自噬提高临床肿瘤放疗疗效提供思路。Abstract: Radiotherapy is one of the most common strategies to treat tumor. But tumor can extremely reduce the effect of therapy due to radioresistance, contributing to the failure of tumor treatment and the recurrence and metastasis of tumor. As a catabolic process common in cells, autophagy plays a vital role in maintaining intracellular homeostasis. Studies have revealed that autophagy has both accelerative and suppressible functions on tumor. On the one hand, autophagy enables tumor to adapt to stress environment such as hypoxia and innutrition, which enhances the abilities of tolerance and viability. On the other hand, excessive autophagy can cause the self-degradation of tumor cells in a long term and resulting in cell death. In this article, according to research progress in recent years, the specific mechanism of autophagy regulating tumor radiosensitivity was reviewed, so as to provide new thoughts to improve the efficacy of radiotherapy by targeted autophagy.
