Abstract:
Objective To observe the influence of memantine on ropivacaine - induced convulsion in mice. Methods Mice were randomly divided into four groups : ropivacaine (R) group, memantine 5 mg·kg-1 (Mem5) group , 10 mg·kg-1 (Mem10) group and 20 mg·kg-1 (Mem20) group. Normal saline was administered intragastrically in R group. Corresponding dose of memantine was administered intragastrically in Mem5, Mem10 and Mem 20 group . 30 minutes later 89.4 mg·kg -1 ropivacaine was injected intraperitoneally in each group. The convulsion latency, convulsion duration and convulsion level of mice were observed. Results Compared with R group, the convulsion latency in Mem5 and Mem10 group extended (P < 0.05, P < 0.01), the convulsion duration in Mem5 and Mem10 group shortened significantly (P < 0.01), and the convulsion level in Mem10 group reduced. The convulsion latency in Mem20 group shortened (P < 0.05), the convulsion duration in Mem20 group extended (P < 0.05). Conclusion In the range of 5 mg·kg-1 to 10 mg·kg-1 memantine can antagonize ropivacaine - induced convulsion, while 20 mg·kg-1 memantine can aggravate ropivacaine - induced convulsion in mice. convulsion latency in Mem5 and Mem 10 group extended (P < 0.05, P < 0.01), the convulsion duration in Mem5 and Mem 10 group shortened significantly (P < 0.01), and the convulsion level in Mem10 group reduced .
