Abstract:
Objective: In order to?find the relationship between phosphodiesterase 4D interacting protein(PDE4DIP) gene and Budd-Chiari syndrome(BCS). Methods: The small interference RNA(siRNA) targeting PDE4DIP gene and the controls were transfected into human umbilical vein endothelia cells(HUVECs), respectively. Then the CCK8 assay, migration scratch assay and tube formation assay were applied to detect the activity of transfected cells on cell proliferation, migration and angiogenesis. Results: The qRT-PCR and western blot assays revealed that the silencing of PDE4DIP gene?in HUVECs cells were both decreased in mRNA or protein levels with P<0.05, respectively. After silencing the PDE4DIP gene, the activity of cell proliferation of HUVECs cells did not change significantly(P=0.51). The silencing of PDE4DIP gene could decrease the ability of migration and angiogenesis, after comparing with the controls, and the difference was statistically significant(P=0.003, P=0.001). Conclusions: The silencing of PDE4DIP gene could inhibit the ability of migration and angiogenesis in HUVECs cells, which may be associated with the membrane formation of BCS.
