Abstract:
Objective: To explore the molecular mechanism of TSG101 regulating the proliferation of hepatoma cells. Methods: In HepG2 and SMMC-7721 cells, constructed the stable expression of TSG101 shRNA cells lines and transient transfection of exogenous TSG101. 1. Western blotting and immunohistochemistry were used to analyze the expression of TSG101 in HCC; 2. EdU, cell colony, CCK-8 were used to analyze the effect of TSG101 on the proliferation of hepatoma cells; 3. Western blotting was used to analyze the molecular mechanisms of TSG101 regulating the proliferation of hepatoma cells. Results: 1. The expression of TSG101 is increased in HCC; 2. TSG101 can promote the proliferation of hepatoma cells; 3. TSG101 can negatively regulate the expression of p53. Conclusion: 1 . TSG101 is a cancer promoting factor;2. TSG101 can promote the proliferation hepatoma cells by negatively regulating p53
