    Rab1A对胶质瘤增殖及其机制的研究

    Study on the proliferation of glioma and its related mechanism by Rab1A

    • 摘要: 目的 探讨Rab1A基因对胶质瘤U251和U87细胞增殖能力的影响及其相关机制。方法 1.通过siLentFect分别将Negative control siRNA、Rab1A siRNA转染入胶质瘤细胞;2.采用Western blot法检测转染前、后胶质瘤细胞Rab1A蛋白的表达;3.应用CCK-8细胞增殖实验和EdU实验检测Rab1A siRNA对胶质瘤U251和U87细胞增殖的影响;4.采用Western blot法分析Rab1A影响胶质瘤增殖的分子机制。结果 1.下调Rab1A基因的表达对胶质瘤U251和U87细胞增殖能力具有明显抑制作用;2.下调Rab1A基因的表达对胶质瘤U251和U87细胞增殖能力具有明显抑制作用;3.Rab1A能够正调控P70S6K。结论 1.Rab1A siRNA能够有效抑制胶质瘤U251和U87细胞中Rab1A蛋白的表达。2.Rab1A基因可能通过正调控P70S6K促进胶质瘤U251和U87细胞增殖能力。

       

      Abstract: Objectives To investigate the effect of Rab1A gene on the proliferation of glioma U251 and U87 cells and its related mechanisms.Methods 1.Negative control siRNA and Rab1A siRNA were transfected into glioma cells by siLentFect.2.Western blot was used to detect the expression of Rab1A protein in glioma cells before and after transfection.3.CCK-8 assay and EdU assay were used to detect the effect of Rab1A siRNA on the proliferation of U251 and U87 cells.4. The molecular mechanism of Rab1A affecting glioma proliferation was analyzed by Western blot.Results 1.The expression of Rab1A gene can inhibit the proliferation of glioma U251 and U87 cells. 2. down regulation of Rab1A gene expression can inhibit the proliferation of glioma U251 and U87 cells. 3.Rab1A can regulate P70S6K.Conclusions 1. Rab1A siRNA can effectively inhibit the expression of Rab1A protein in glioma U251 and U87 cells.2.Rab1A gene may promote the proliferation of glioma U251 and U87 cells by regulating P70S6K.
