    小鼠骨髓源嗜酸性粒细胞的体外诱导与鉴定

    Differentiation and identification of eosinophil derived from bone marrow cells of Mice

    • 摘要: 目的 分离小鼠骨髓细胞并将其诱导为嗜酸性粒细胞,为后续嗜酸性粒细胞功能探讨奠定基础。方法 剖杀小鼠,分离其股骨骨髓细胞,用rmIL-5等对其进行定向诱导分化,用瑞氏染色及Chromotrope 2R染色法对所诱导细胞进行染色观察并利用流式细胞术对其表面CCR3(C-C Motif Chemokine Receptor 3)/Siglec-F(Sialic Acid Binding Ig Like Lectin 5)等分子表达予以检测,从细胞形态学及表面分子表达情况对所诱导的细胞予以鉴定。 结果 瑞氏染色及Chromotrope 2R染色可见嗜酸性粒细胞典型形态,流式细胞术检测Siglec-F及CCR3表达水平明显升高。结论 本研究利用rmIL-5在体外成功获得较高纯度的小鼠骨髓来源的嗜酸性粒细胞。

       

      Abstract: In order to eosinophil in vitro, bone marrow cells from BALB/c mice were induced to differentiate the eosinophil using the rmIL-5. Typical eosinophil granulocyte were observed in the differentiated cells by using the Wright’s staining and Chromotrope 2R staining. The expression of specific markers of EOS-CCR3 (c-c Motif Chemokine Receptor 3) and Siglec-F (Sialic Acid Binding Ig Like Lectin 5) were detected flow cytometry, and the data showed that the level of both CCR3 and Siglec-F were significantly increased (P<0.001). This study confirmed that the high purity of eosinophil granulocyte can be induced by the rmIL-5 in the bone marrow cells from the BALB/c mice in vitro.
