    深裂竹根七乙酸乙酯提取物中的化学成分及抗肿瘤活性研究

    The study on the antitumor effects and chemical components of the ethyl acetate extracts from Disporposis pernyi

    • 摘要: 目的 研究深裂竹根七乙酸乙酯提取物中化合物的种类与抗肿瘤活性。方法 采用系统溶剂法和柱色谱法提取分离深裂竹根七乙酯乙酯提取物中的化合物,用核磁共振、质谱和薄层方法鉴别其分子结构。采用MTT法测定提取物对人源乳腺癌瘤株MCF-7的抑制活性,用AO-EB荧光双染法观察细胞形态学变化。结果 从深裂竹根七乙酸乙酯提取物中分离并鉴别了3种成分,分别为白桦脂醇、β-胡萝卜苷和木犀草素,其中白桦脂醇为首次从深裂竹根七中分离得到。提取物对MCF-7细胞增殖有较强的抑制作用。与空白组相比,药物组细胞死亡明显增多。结论 深裂竹根七乙酸乙酯提取物对人源乳腺癌瘤株MCF-7具有抑制活性,其中白桦脂醇等成分起到一定作用。

       

      Abstract: Objective To investigate the chemical constituents and pharmacological activity of the ethyl acetate extract fromDisporposis pernyi. Methods Compounds in the ethyl acetate extract of Disporposis pernyiwere extracted and separated by systematic solvent extraction and column chromatography. The molecular structures were identified by nuclear magnetic resonance (NMR), mass spectrometry (MS) and thin layer chromatography (TLC). The inhibitory activity of extracts against human breast cancer cell line MCF-7 was determined by MTT method. Cell morphology was observed with AO-EB fluorescent staining . Results Three components were isolated and identified, which were betulin, β-daucosterol and luteolin. Betulin was first isolated from Disporposis pernyi. The extract has inhibitory effects on the proliferation of MCF-7 cells. Compared with the control group, the number of apoptotic cells of the drug group increased significantly. Conclusions The ethyl acetate extract from Disporposis pernyi has an inhibitory activity on human breast cancer cell MCF-7, among which betulin plays a certain role.
