    The effect of DKC1 gene on the proliferation and metastasis of colorectal cancer cells

    • Objective To investigate the effect of dyskerin pseudouridine synthase 1(DKC1)on the proliferation and metastasis of colorectal cancer cells. Methods Human colorectal cancer cells HCT116 and DLD1 were transiently transfected with Control-siRNA(siCtrl group)and DKC1-siRNA(siDKC1 group),and the knockdown effect of DKC1 protein level was verified by Western blot.The effect of DKC1 on the proliferation and metastasis ability of colorectal cancer cells HCT116 and DLD1 was detected by CCK-8 assay,wound healing assay and Transwell assay. Results The results of Western blot showed that the level of DKC1 protein was significantly decreased after transfection of DKC1-siRNA.The results of CCK-8 assay showed that the proliferation ability of colorectal cancer cells HCT116 and DLD1 were significantly decreased after transfection of DKC1-siRNA(P<0.01).Wound healing assay and Transwell assay showed that the metastasis ability of colorectal cancer cells HCT116 and DLD1 were significantly decreased after transfection of DKC1-siRNA(P0.05).Conclusions Knockdown of DKC1 can inhibit the proliferation and metastasis of colorectal cancer cells.
