Effects of PAK4 inhibitor on the migration,invasion and proliferation of glioma cells

Abstract
Objective To observe the effects of p21-activated kinase 4 (PAK4) inhibitor on the invasion, migration and proliferation of glioma cells. Methods Cell lines with high expression of PAK1 protein were screened out by Western blot and the level of PAK4 in glioma cell lines was detected after treatment with PAK4 inhibitor.The 50% inhibitory concentration (IC50) of PAK4 inhibitor on U87 cells and T98G cells was determined by CCK 8 assay.The difference of migration and invasion abilities between the experimental and control groups was compared by Transwell assay and wound healing test. Flow cytometry was used to detect the effect of PAK4 inhibitor on the apoptosis of U87 and T98G cells. Colony formation assay was used to detect the effect of PAK4 inhibitor on the proliferation of U87 and T98G cells. Results PAK4 protein was highly expressed in U87 and T98G cells.The IC50 was 1.61 mg/L for U87 cells and 1.52 mg/L for T98G cells.In the PAK4 inhibitor group, the number of U87 and T98G cells entering into the lower chamber of Transwell significantly reduced. The wound healing rate remarkably reduced for cells in the PAK4 inhibitor group. The proliferation of U87 and T98G cells in the PAK4 inhibitor group significantly reduced, with an increased number of apoptotic cells. PAK4 inhibitor remarkably suppressed the proliferation of glioma U87 and T98G cells. Conclusions PAK4 inhibitor can weaken the invasion, migration and proliferation abilities of glioma cells.
