The expression and mechanism of circular RNA hsa_circ_0081069 in colorectal cancer

Abstract
Objective To explore the expression of circular RNA hsa_circ_0081069 in colorectal cancer and its effect on the migration, invasion and proliferation of colorectal cancer cells. Methods The levels of hsa_circ_0081069 in colorectal cancer tissues and its adjacent tissue were detected by qRT-PCR. The effects of hsa_circ_0081069 knockdown on the migration, invasion and proliferation of colorectal cancer cells were measured by Transwell assay and cell counting kit-8 assay. Furthermore, the dual-luciferase reporter experiment was used to detect the interaction between hsa_circ_0081069 and miR-1205. Results Compared with adjacent normal tissues, the levels of hsa_circ_0081069 in colorectal cancer tissues significantly increased (P<0.001). Compared with human normal colorectal mucosa cell line FHC, hsa_circ_0081069 was significantly up-regulated in six colorectal cancer cell lines (P<0.05), especially in HCT116 and DLD1 cells (P<0.001). Knockdown of hsa_circ_0081069 inhibited the migration, invasion and proliferation of HCT116 and DLD1 cells (P<0.01). Furthermore, hsa_circ_0081069 targeted miR-1205, while inhibiting miR-1205 after silencing hsa_circ_0081069 reversed the inhibited migration, invasion and proliferation of colorectal cancer cells. Conclusions In the study, hsa_circ_0081069 is highly expressed in colorectal cancer tissues and cells, and promotes the migration, invasion and proliferation of colorectal cancer through targeting miR-1205.
