The role and mechanism of CPEB3 in glycolytic metabolism of breast cancer cells

Abstract
Objective To investigate the role and mechanism of CPEB3 in the glycolytic metabolism of breast cancer cells. Methods The expression of CPEB3 was detected by RT-qPCR and Western blot. The proliferation of MCF-7 cells was assessed by CCK-8 assay. The cell apoptosis was measured by Annexin V-FITC/PI kit. The migration of the cells was evaluated by Transwell assay. The glucose concentration and lactate content in culture medium were determined by colorimetric assay kits. Results CPEB3 was significantly down-regulated in breast cancer cells, compared with normal breast epithelial cells (P<0.05). Overexpression of CPEB3 markedly inhibited the proliferation, migration and glycolysis of MCF-7 cells (P<0.05), while promoting their apoptosis (P<0.05). Moreover, CPEB3 overexpression obviously reduced the expression of IL-6R and p-STAT3 (P<0.05), while STAT3 overexpression significantly reversed the inhibitory effect of CPEB3 on glycolysis in MCF-7 cells (P<0.05). Conclusions CPEB3 inhibits glycolysis in breast cancer cells through regulating the IL-6R/STAT3 pathway.
