Expression of miR-146b-5p and TRIM29 in endometrial carcinoma

Abstract
Objective To investigate the expression of miRNA-146b-5p(miR-146b-5p) and tripartite motif protein 29(TRIM29) in endometrial cancer, and its clinical significance.Methods A total of 60 patients with endometrial cancer and 30 patients with normal proliferative endometrium were selected from the Affiliated Hospital of Xuzhou Medical University and the Second Affiliated Hospital of Xuzhou Medical University from October 2019 to October 2020. Their endometrial tissues were collected to detect the expression of TRIM29 by immunohistochemistry. The expression of miR-146b-5p and TRIM29 mRNA was measured by qRT-PCR. The correlation between the expression of miR-146b-5p and TRIM29 was evaluated by Spearman analysis. Then, Kaplan-Meier method and Cox proportional risk regression model were used to analyze the prognosis.Results According to immunohistochemical results, the positive expression rate of TRIM29 in endometrial carcinoma tissues(55.0%) was lower than that in normal proliferative endometrial tissues(76.7%)(P<0.05). Then, qRT-PCR results showed that the expression of miR-146b-5p in endometrial cancer tissues was significantly higher than that in normal proliferative endometrial tissues, while the expression of TRIM29 was significantly lower than that in normal proliferative endometrial tissues(P<0.05). Spearman analysis indicated that the expression of miR-146b-5p was negatively correlated with TRIM29(r=-0.595, P<0.05). Kaplan-Meier analysis showed that patients with high expression of miR-146b-5p and low expression of TRIM29 had poor prognosis(P<0.05). Cox multivariate regression analysis demonstrated that miR-146b-5p and TRIM29 were the independent factors affecting the prognosis of patients with endometrial cancer.Conclusions High expression of miR-146b-5p and low expression of TRIM29 in endometrial cancer tissues may be involved in the pathogenesis of endometrial cancer.
