The role of DKC1 gene in proliferation and migration of human renal carcinoma cells

Abstract
Objective To investigate the effects of DKC1 knockdown by small interfering RNA(siRNA) on the proliferation and migration of human renal carcinoma ACHN cells and 786-O cells. Methods The scrambled siRNA and DKC1 siRNA fragments were transiently transfected into ACHN and 786-O cells. The level of DKC1 and EZH2 were detected by Western blot. The effects of DKC1 knockdown on cell proliferation were tested by cell proliferation assay. The effects of DKC1 knockdown on the migration of ACHN and 786-O cells were explored by Transwell assay. Results DKC1 siRNA could down-regulated the level of DKC1 in ACHN and 786-O cells and inhibited the capacity of proliferation and migration of cells via reducing EZH2 expression. Conclusion DKC1 knockdown can reduce the expression of EZH2, which would then inhibit the proliferation and migration of renal carcinoma cells
