Abstract
Objectives To investigate the effect of Rab1A gene on the proliferation of glioma U251 and U87 cells and its related mechanisms.Methods 1.Negative control siRNA and Rab1A siRNA were transfected into glioma cells by siLentFect.2.Western blot was used to detect the expression of Rab1A protein in glioma cells before and after transfection.3.CCK-8 assay and EdU assay were used to detect the effect of Rab1A siRNA on the proliferation of U251 and U87 cells.4. The molecular mechanism of Rab1A affecting glioma proliferation was analyzed by Western blot.Results 1.The expression of Rab1A gene can inhibit the proliferation of glioma U251 and U87 cells. 2. down regulation of Rab1A gene expression can inhibit the proliferation of glioma U251 and U87 cells. 3.Rab1A can regulate P70S6K.Conclusions 1. Rab1A siRNA can effectively inhibit the expression of Rab1A protein in glioma U251 and U87 cells.2.Rab1A gene may promote the proliferation of glioma U251 and U87 cells by regulating P70S6K.
