Differentiation and identification of eosinophil derived from bone marrow cells of Mice

Abstract
In order to eosinophil in vitro, bone marrow cells from BALB/c mice were induced to differentiate the eosinophil using the rmIL-5. Typical eosinophil granulocyte were observed in the differentiated cells by using the Wright’s staining and Chromotrope 2R staining. The expression of specific markers of EOS-CCR3 (c-c Motif Chemokine Receptor 3) and Siglec-F (Sialic Acid Binding Ig Like Lectin 5) were detected flow cytometry, and the data showed that the level of both CCR3 and Siglec-F were significantly increased (P<0.001). This study confirmed that the high purity of eosinophil granulocyte can be induced by the rmIL-5 in the bone marrow cells from the BALB/c mice in vitro.
