GOLPH3 shRNA decreases migration and invasion of human glioma

Abstract
objective To investigate the effect and mechanism of GOLPH3 on the human glioma cells migration and invasion.Methods Transfection of the GOLPH3 shRNA with GFP tagged plasmid into the U25l glioma cell were carried out using Lipofectamine 2000.Wound healing assay and Transwell invasion assay were used to analyze the influence on the migration and invasion of glioma U251 cells after transfection of the GOLPH3 shRNA with GFP tagged plasmid. Western blot was used to analyze the protein expressions of YB1/ P-YB1 after GOLPH3 down-regulation . Results Compared to the control plasmids, GOLPH3 shRNA with GFP tagged plasmid could decrease the expression of YB1/ P-YB1 in glioma U251 cells . In cell invasion assay, GOLPH3 shRNA inhibits cell invasion ability by 49%, In cell migration assay,GOLPH3 shRNA inhibits cell migration ability by 58%; Down-regulation of GOLPH3 decreased the levels of YB-1 and pYB-1S102 in glioma U251 cells. Conclusion Down-regulation of GOLPH3 may be inhibit glioma cells migration and invasion via decreasing the expression of YB1/ P-YB1
